Frankia Defined Propionate Minimal Medium (DPM)
For routine growth and subculture of Frankia
| Component | Per liter |
| NaPropionate | 1.2 g |
| KH2PO4 | 1.0 g |
| MgSO4.7H2O | 0.1 g |
| CaCl2.2H2O | 0.01 |
| FeSO4.7H2O/EDTA stock | 1.0 ml |
| Hoagland's microelements | 1.0 ml |
| (adjust pH to 6.5) | |
This medium was used initially as N-deficient medium for isolating Frankia from nodules and soil. Ammonium chloride can be added (0.5 g/l) to make it complete (DPM+N) for growth of Frankia strains in culture.
1000X Stock Iron: Dissolve 0.75 g Na2EDTA.2H2O, 0.56 g FeSO4.7H2O ( make up to 100 ml).
Hoagland's Microelements:
| Component | (g/l) |
| H3BO4 | 2.8 |
| MnCl2.4H20 | 1.8 |
| ZnSO4.7H2O | 0.2 |
| CuSO4.5H2O | 0.1 |
| NaMoO4 | 0.025 |
Notes:
- Proteose Peptone #3 can be added at a level of 0.5 g/l for some growth improvement.
- Although more expensive, MOPS can be added to better control pH. Make up a 10X NaMOPS solution to 200 mM, pH 6.5 by adding 46 g to 1 liter dH2O and then add at a rate of 100 ml per liter, keeping all other weighed components the same.
